Basic differential backup questions

I have a 2TB external hard drive that got corrupted and was no longer usable. I copied files from my back-up drive (used solely for back-up) to a new external drive. I'm trying to determine how to do differential back-up from my new external drive to my back-up drive. Can I do this without doing a full back-up from my new external drive to my back-up drive (used solely for back-up) since the back-up drive is where I got the files from to put in my external drive to begin with? And, if so, how do I do it? Sorry if these are inane questions. I just downloaded the free AOMEI software and haven't used this before so I might be missing something obvious.
